In this communication, the authors report the further optimization of I by conformational restraint of the side chain which has led to more potent and selective compounds, exemplified by II. II was evaluated in a functional model of 5-HT7 receptor activation by examination of adenylyl cyclase activity in HEK 293 cells stably expressing the human 5-HT7(a) receptor.
