3034-53-5, 2-Bromothiazole (13.0 g, 1.0 eq), 1-methyl-imidazole (2.0 eq), Cul (0.05 eq) and K4[Fe(CN)6] (0.1 eq) were combined in 80 mL of dry NMP and heated in a sealed tube at 140 C for 16h. To a cooled solution (-78°C) of n-butyllithium (1.7M in pentane, 323mL,550.8 mmol, 1.5 eq) in tetrahydrofuran (150 mL) under nitrogen was added 2- bromothiazole (60 g, 367.2mmol). The resulting suspension was stirred below – 78°C for 45 min. The lithiated thiazole solution was transferred via cannula to a solution of dimethylformamide (25 mL) in tetrahydrofuran (200 mL) at -78°C. The reaction was allowed to warm to room temperature over 3 h and quenched by the addition of water (100 mL). The aqueous phase was extracted with ethyl acetate (3*100 mL). Combined organic extracts were dried over anhydrous magnesium sulfate, filtered and reduced to afford 2-formylthiazole as a crude oil.
