Du, Zhi; Yu, Dongqin; Du, Xiubo; Scott, Peter; Ren, Jinsong; Qu, Xiaogang published an article in 2019, the title of the article was Self-triggered click reaction in an Alzheimer’s disease model: in situ bifunctional drug synthesis catalyzed by neurotoxic copper accumulated in amyloid-beta plaques.COA of Formula: C14H12N2S And the article contains the following content:

Cu is one of the essential elements for life. Its dyshomeostasis has been demonstrated to be closely related to neurodegenerative disorders, such as Alzheimer’s disease (AD), which is characterized by amyloid-beta (Abeta) aggregation and Cu accumulation. It is a great challenge as to how to take advantage of neurotoxic Cu to fight disease and make it helpful. Herein, we report that the accumulated Cu in Abeta plaques can effectively catalyze an azide-alkyne bioorthogonal cycloaddition reaction for fluorophore activation and drug synthesis in living cells, a transgenic AD model of Caenorhabditis elegans CL2006, and brain slices of triple transgenic AD mice. More importantly, the in situ synthesized bifunctional drug 6 can disassemble Abeta-Cu aggregates by extracting Cu and photo-oxygenating Abeta synergistically, suppressing Abeta-mediated paralysis and diminishing the locomotion defects of the AD model CL2006 strain. Our results demonstrate that taking the accumulated Cu ions in the Abeta plaque for an in situ click reaction can achieve both a self-triggered and self-regulated drug synthesis for AD therapy. To the best of our knowledge, a click reaction catalyzed by local Cu in a physiol. environment has not been reported. This work may open up a new avenue for in situ multifunctional drug synthesis by using endogenous neurotoxic metal ions for the treatment of neurodegenerative diseases. On February 23, 2005, Solbach, C.; Uebele, M.; Reischl, G.; Machulla, H.-J. published an article.Quality Control of 2-(4-Aminophenyl)-6-methylbenzothiazole The title of the article was Efficient radiosynthesis of carbon-11 labelled uncharged Thioflavin T derivatives using [11C]methyl triflate for β-amyloid imaging in Alzheimer’s Disease with PET. And the article contained the following:

The synthesis of carbon-11 amino function labeled uncharged Thioflavin T derivatives is known to be performed by reaction of the demethyl-precursors with [11C]methyl iodide but the labeling yields are only mediocre. The use of [11C]methyl triflate improved the radiochem. yield of three potential β-amyloid imaging PET-radiotracers significantly. Performance of the labeling reaction by reacting the corresponding precursor mols. with [11C]methyl triflate for 1 min at 80° led to radiochem. yields of 44±10% (n = 5) for [11C]6-Me-BTA-1, 68±4% (n = 10) for [11C]BTA-1 and 58±2% (n = 5) for [11C]6-OH-BTA-1 with respect to [11C]methyl triflate. In production runs (60 min, 50 μA) up to 6500 MBq (mean: 4000±1900 MBq) of [11C]6-Me-BTA-1, 7900 MBq (mean: 6000±1000 MBq) of [11C]BTA-1 and 7100 MBq (mean: 6300±600 MBq) of [11C]6-OH-BTA-1 could be obtained ready for i.v. injection. The radiochem. purity was >95% with specific activities in the range of 80-120 GBq/μmol (EOS) within a total synthesis time of less than 40 min after EOB. And the article contained the following:

G-quadruplex has attracted considerable attention due to their prevalent distribution in functional genomic regions and transcripts, which can importantly influence biol. processes such as regulation of telomere maintenance, gene transcription and gene translation. Artificial receptor study has been developed for accurate identification of G-quadruplex from DNA species, since it is important for the G-quadruplex related basic research, clin. diagnosis, and therapy. Herein, fluorescent dye ThT-E, a derivative of the known fluorescence probe Thioflavin T (ThT), was designed and synthesized to effectively differentiate various G-quadruplex structures from other nucleic acid forms. Compared with Me groups in ThT, three Et groups were introduced to ThT-E, which leads to strengthened affinity, selectivity and little inducing effect on the G-quadruplex formation. More importantly, ThT-E could be served as a visual tool to directly differentiate G-quadruplex solution even with naked eyes under illumination of UV light. Thus, this probe reported herein may hold great promise for high-throughput assay to screen G-quadruplex, which may widely apply to G-quadruplex-based potential diagnosis and therapy. SSAs are a class of supramol. self-associating amphiphilic salt, the anionic component of which contains a covalently bound hydrogen bond donor-acceptor motif. This results in a monomeric unit which can adopt multiple hydrogen bonding modes simultaneously. Previous investigations have shown examples of SSAs to act as antimicrobial agents against clin. relevant methicillin-resistant Staphylococcus aureus (MRSA). Herein, we report an intrinsically fluorescent SSA which can self-associate producing dimers, spherical aggregates and hydrogels dependent on solvent environment, while retaining antimicrobial activity against both model Gram-pos. (MRSA) and Gram-neg. (Escherichia coli) bacteria. Finally, we demonstrate the SSA supramol. hydrogel to tolerate the inclusion of the antibiotic ampicillin, leading to the enhanced inhibition of growth with both model bacteria, and derive initial mol. structure-physicochem. property-antimicrobial activity relationships. The Article related to aqueous solubility model druglike analysis, Pharmacology: Methods and other aspects.Quality Control of 2-(4-Aminophenyl)-6-methylbenzothiazole

On August 31, 2007, Wang, Junmei; Krudy, George; Hou, Tingjun; Zhang, Wei; Holland, George; Xu, Xiaojie published an article.Quality Control of 2-(4-Aminophenyl)-6-methylbenzothiazole The title of the article was Development of Reliable Aqueous Solubility Models and Their Application in Druglike Analysis. And the article contained the following:

In this work, two reliable aqueous solubility models, ASMS (aqueous solubility based on mol. surface) and ASMS-LOGP (aqueous solubility based on mol. surface using calculated log P (ClogP) as a descriptor), were constructed by using atom type classified solvent accessible surface areas and several mol. descriptors for a diverse data set of 1708 mols. For ASMS (without using ClogP as a descriptor), the leave-one-out q2 and root-mean-square error (RMSE) were 0.872 and 0.748 log unit, resp. ASMS-LOGP was slightly better than ASMS (q2 = 0.886, RMSE = 0.705). Both models were extensively validated by three cross-validation tests and encouraging predictability was achieved. High throughput aqueous solubility prediction was conducted for a number of data sets extracted from several widely used databases. The authors found that real drugs are about 20-fold more soluble than the so-called druglike mols. in the ZINC database, which have no violation of Lipinski’s “Rule of 5” at all. Specifically, oral drugs are about 16-fold more soluble, while injection drugs are 50-60-fold more soluble If the criterion of a mol. to be soluble is set to -5 log unit, about 85% of real drugs are predicted as soluble; in contrast only 50% of druglike mols. in ZINC are soluble The authors concluded that the two models could be served as a rule in druglike anal. and an efficient filter in prioritizing compound libraries prior to high throughput screenings (HTS). And the article contained the following:

OBJECTIVE: To grasp the endemic situation of schistosomiasis in Jingzhou City in 2017, so as to provide the evidence for formulating prevention strategies. METHODS: According to the requirements of National Schistosomiasis Surveillance Program (2014), Hubei Schistosomiasis Surveillance Program (2015) and Jingzhou Schistosomiasis Surveillance Program, the local population infection monitoring, mobile population infection monitoring, livestock disease monitoring, snail monitoring, and wild manure monitoring were carried out at 71 monitoring sites throughout the city. RESULTS: Among the 71 surveillance sites, 44 118 local residents received the indirect hemagglutination test (IHA) for schistosomiasis and 1 925 persons were positive, with the positive rate of 4.36%, However, no stool examination positive cases were found. Totally 2 175 mobile people received the IHA for schistosomiasis and 93 persons were positive, with the positive rate of 4.28%, but no stool examination positive cases were found. A total of 1 937 head of cattle received the stool examination for schistosome infection, but no positives were found. Totally 1 302 Oncomelania hupensis habitats were found in 69 surveillance sites, with 1 923.64 hm2 area with snails. Totally 348 756 frames were surveyed, and the occurrence rate of snails was 17.19%. A total of 142 494 living snails were captured, with the average density of 0.41 snail/0.1 m2. No schistosome-infected snails were found. Totally 596 cases of wild manure were picked up in 41 environments with snails among 23 surveillance sites, but schistosome infested wild manure was not found. CONCLUSIONS: In 2017, the level of schistosomiasis epidemic was very low in Jingzhou City. However, the risk of epidemic rebound still exists, and therefore, it is necessary to strengthen the monitoring work to further consolidate the achievements of schistosomiasis control. And the article contained the following:

BACKGROUND: Body mass index (BMI) is a significant predictor of developing end-stage renal disease (ESRD). The relation between a change in BMI (DeltaBMI) and the incidence of ESRD has not been examined in any large epidemiologic studies. METHODS: We determined the DeltaBMI in subjects who participated in the Okinawa General Health Maintenance Association (OGHMA) screenings in 1983 and again in 1993. Screenees were free of ESRD at the 1993 screening and were then monitored until the end of 2000 to determine whether they developed ESRD. Participants were identified using ID numbers, birthdates, and other identifiers. Details of every ESRD patient treated in Okinawa are maintained in an independent community-based dialysis registry. Multivariate logistic analyses were performed to determine the significance of a DeltaBMI on the incidence of ESRD using SAS. The ethics committee of the OGHMA approved the study protocol. Only coded data were used for this study. RESULTS: Among the 92,364 subjects aged 30-89 years screened in 1983, 29,011 (31.4%) returned for the 1993 screening. The median DeltaBMI was 2.1%, and the subjects were divided into two groups: DeltaBMI < 2.1% (G1) and DeltaBMI > or = 2.1% (G2). The cumulative incidence of ESRD was 0.31% in G1 (ESRD in 44) and 0.14% in G2 (ESRD in 21). The odds ratio (95% confidence interval) of developing ESRD based on a DeltaBMI was 2.268 (1.284-4.000, P < 0.01) after adjusting for age, sex, systolic blood pressure, BMI in 1983, and proteinuria. CONCLUSION: The findings of the present study suggest that a DeltaBMI is an independent risk factor for the incidence of ESRD, especially for those with proteinuria. The reasons for the BMI change were not recorded in this study. Unintentional weight loss, however, might warrant evaluation for the presence or progression of chronic kidney disease. On February 2, 2012, Orellana, M Angeles; Gómez-Lus, M Luisa; Lora, David published an article.Computed Properties of 92-36-4 The title of the article was Sensitivity of Gram stain in the diagnosis of urethritis in men.. And the article contained the following:

BACKGROUND: Acute urethritis is among the most common types of sexually transmitted diseases in men. The diagnosis usually requires microscopic evidence of urethritis, but sometimes urethral pathogens are detected in asymptomatic men without such evidence. The aims of this study were to assess the sensitivity of Gram stain in men with urethral symptoms and to relate it to the microorganisms isolated. METHODS: Between January 2006 and December 2007, 491 urethral samples were analysed. The authors assessed the presence of leukocytes by Gram stain and tested specifically for Chlamydia trachomatis, Ureaplasma urealyticum, Mycoplasma hominis and Trichomonas vaginalis, as well as analysing the results of conventional culture. RESULTS: The percentages of positive samples as a function of Gram category were two or less polymorphonuclear leukocytes (PMNLs)/high-power field (HPF) 25% (92/364), three to four PMNLs/HPF 32% (18/57) and five or more PMNLs/HPF 54% (38/70). Classing samples with more than two PMNLs/HPF as positive, the sensitivity, specificity and positive likelihood ratio for Gram stain were 38% (95% CI 30 to 46), 79% (95% CI 75 to 84) and 1.8 (95% CI 1.4 to 2.4), respectively. On the other hand, taking as positive five or more PMNLs/HPF, the sensitivity, specificity and positive likelihood ratio for Gram stain were 26% (95% CI 18 to 33), 91% (95% CI 87 to 94) and 2.7 (95% CI 1.8 to 4.2), respectively. The sensitivity of Gram stain to Neisseria gonorrhoeae, Chlamydia trachomatis and Ureaplasma urealyticum were 80% (95% CI 64 to 96), 23% (95% CI 8 to 39) and 11% (95% CI 2 to 20), respectively. CONCLUSION: The low sensitivity of Gram stain means that negative results do not exclude the presence of urethritis in symptomatic patients. And the article contained the following:

OBJECTIVE: Given that intention to breastfeed is a strong predictor of breastfeeding initiation and duration, the objectives of this study were to estimate the population-based prevalence and the factors associated with the intention to breastfeed. DESIGN: Retrospective population-based cohort study. SETTING: All hospitals in Ontario, Canada (1 April 2009-31 March 2010). POPULATION: Women who gave birth to live, term, singletons/twins. METHODS: Patient, healthcare provider, and hospital factors that may be associated with intention to breastfeed were analysed using univariable and multivariable regression. MAIN OUTCOME MEASURES: Population-based prevalence of intention to breastfeed and its associated factors. RESULTS: The study included 92,364 women, of whom 78,806 (85.3%) intended to breastfeed. The odds of intending to breastfeed were higher amongst older women with no health problems and women who were cared for exclusively by midwives (adjusted OR 3.64, 95% CI 3.13-4.23). Being pregnant with twins (adjusted OR 0.73, 95% CI 0.57-0.94), not attending antenatal classes (adjusted OR 0.58, 95% CI 0.54-0.62), having previous term or preterm births (adjusted OR 0.79, 95% CI 0.78-0.81, and adjusted OR 0.87, 95% CI 0.82-0.93, respectively), and delivering in a level-1 hospital (adjusted OR 0.85, 95% CI 0.77-0.93) were associated with a lower intention to breastfeed. CONCLUSIONS: In this population-based study ~85% of women intended to breastfeed their babies. Key factors that are associated with the intention to breastfeed were identified, which can now be targeted for intervention programmes aimed at increasing the prevalence of breastfeeding and improving overall child and maternal health. And the article contained the following:

OBJECTIVE: Because breastfeeding is the optimal form of infant feeding, this study was conducted to determine the effect of gestational age on breastfeeding in term infants. STUDY DESIGN: A retrospective population-based cohort study of singleton/twin hospital births was conducted in Ontario, Canada between April 1, 2009, and March 31, 2010. Multivariate logistic regression was used to determine the adjusted effect of gestational age on breastfeeding. RESULTS: Our study population comprised 92,364 infants, of whom 80,297 (86.9%) were exclusively or partially breastfed at the time of hospital discharge. Multivariate logistic regression analyses demonstrated that early-term infants had lower odds of being breastfed compared with infants born at 41 weeks gestation (40 weeks: aOR, 0.93; 95% CI, 0.86-0.99; 39 weeks: aOR, 0.87; 95% CI, 0.81-0.93; 38 weeks: aOR, 0.81; 95% CI, 0.75-0.88; 37 weeks: aOR, 0.74; 95% CI, 0.67-0.82). CONCLUSION: Using a population-based approach, we found that infants born at 40, 39, 38, and 37 weeks gestation had increasingly lower odds of being breastfed compared with infants born at 41 weeks. Clinicians need to be made aware of the differences in outcomes of infants delivered at early and late term, so that appropriate breastfeeding support can be provided to women at risk for not breastfeeding.
