A convenient and sensitive colorimetric assay for acetylcholinesterase (AChE) and its inhibitor has been designed based on the oxidase-like activity of {100}-faceted Pd square nanoplates which are grown in situ on reduced graphene oxide (PdSP@rGO). PdSP@rGO can effectively catalyze the oxidation of colorless 3,3′,5,5′-tetramethylbenzidine (TMB) without the assistance of H2O2 to generate blue oxidized TMB (oxTMB) with a characteristic absorption peak at 652 nm. In the presence of AChE, acetylthiocholine (ATCh), a typical AChE substrate, is hydrolyzed to thiocholine (TCh). The generated TCh can effectively inhibit the PdSP@rGO-triggered chromogenic reaction of TMB via cheating with Pd, resulting in color fading and decrease in absorbance. Thus, a sensitive probe for AChE activity is constructed with a working range of 0.25-5 mU mL-1 and a limit of detection (LOD) of 0.0625 mU mL-1. Furthermore, because of the inhibition effect of tacrine on AChE, tacrine is also detected through the colorimetric AChE assay system within the concentrations range 0.025-0.4μM with a LOD of 0.00229μM. Hence, a rapid and facile colorimetric procedure to sensitively detect AChE and its inhibitor can be anticipated through modulating the oxidase-like activity of PdSP@rGO.

Several 2-imino-5-hydroxymethyl-8-methyl-2H-pyrano[2,3-c]pyridine-3-(N-aryl)carboxamides, e.g., I, were prepared by reaction of pyridoxal hydrochloride with various N-aryl cyanoacetamides. Reaction of these compounds with aromatic amines furnished a wide series of 2-(N-R-phenyl) imino-5-hydroxymethyl-8-methyl-2H-pyrano[2,3-c]pyridine-3-carboxamides. Antibacterial and antifungal activities of the synthesized compounds were studied. Most of the obtained compounds demonstrated significant activity against bacterial or fungal strains (MIC in the range of 12.5-25 μg/mL), displaying comparable or even better efficacy than the standard drugs.

A series of new pyrazole linked benzothiazole-β-naphthol derivatives were designed and synthesized using a simple, efficient and ecofriendly route under catalyst-free conditions in good to excellent yields. These derivatives were evaluated for their cytotoxicity on selected human cancer cell lines. Among those, the derivatives 4j, 4k and 4l exhibited considerable cytotoxicity with IC50 values ranging between 4.63 and 5.54 μM against human cervical cancer cells (HeLa). Structure activity relationship was elucidated by varying different substituents on benzothiazoles and pyrazoles. Further, flow cytometric anal. revealed that these derivatives induced cell cycle arrest in G2/M phase and spectroscopic studies such as UV-visible, fluorescence and CD studies showed that these derivatives exhibited good DNA binding affinity. Addnl., these derivatives can effectively inhibit the topoisomerase I activity. Viscosity studies and mol. docking studies demonstrated that the derivatives bind with the minor groove of the DNA.

The effect of increasing concentrations (0, 0.25, 0.5, 1%) of com. water-soluble AstaSana astaxanthin (AST) on the physicochem. properties, release kinetics and antioxidant activity of binary 75/25 CM-cellulose/gelatin (CMC75/GEL25) and octenyl succinic anhydride starch/gelatin (OSA75/GEL25) blend films was investigated. The microscopic observations showed that the obtained blends were phase-separated systems. Regardless of the AST concentration, the CMC-based films were more opaque, stronger, and less stretchable than the OSA-based films. The AST-supplemented films exhibited an intensive red color. The starch granules (from the AST formulation) contributed to high roughness and opacity of the films. The presence of AST contributed to a significant decrease of the puncture strength and oxygen permeability of the CMC-based film. The AST-supplemented films did not differ in terms of the water vapor barrier properties. Due to the low quantity of astaxanthin in the systems, the Fourier-transform IR spectra of the control and AST-supplemented films were similar. The increase in the AST concentration was accompanied by reduced solubility and increased swelling of the OSA-based films, while an opposite result was observed for the CMC-based films. According to the Korsmeyer-Peppas with time lag model, the AST release kinetics exhibited quasi-Fickian behavior. Due to their weaker solubility, the OSA75/GEL25 films offered at least 7 times slower release of AST than the CMC-based carrier. The times required for 25% AST release from the CMC75/GEL25 and OSA75/GEL25 films were 1.20-1.74 and 12.60-20.04 min, resp. A high pos. correlation (R2 = 0.78-0.91) was found between the AST release and antiradical activity of the films.
