By use of iterative design aided by predictive models for target affinity, brain permeability, and hERG activity, novel and diverse compounds based on cyclic amidine and guanidine cores were synthesized with the goal of finding BACE-1 inhibitors as a treatment for Alzheimer’s disease. Since synthesis feasibility had low priority in the design of the cores, an extensive synthesis effort was needed to make the relevant compounds. Syntheses of these compounds are reported, together with physicochemical properties and structure-activity relationships based on in vitro data. Four crystal structures of diverse amidines binding in the active site are deposited and discussed. Inhibitors of BACE-1 with 3 muM to 32 nM potencies in cells are shown, together with data on in vivo brain exposure levels for four compounds. The results presented show the importance of the core structure for the profile of the final compounds.

Discovery of a Pyrimidothiazolodiazepinone as a Potent and Selective Focal Adhesion Kinase (FAK) Inhibitor

Focal adhesion kinase (FAK) is a tyrosine kinase with prominent roles in protein scaffolding, migration, angio-genesis, and anchorage-independent cell survival and is an attractive target for the development of cancer therapeutics. However, current FAK inhibitors display dual kinase inhibition and/or significant activity on several kinases. Although multi-targeted activity is at times therapeutically advantageous, such behavior can also lead to toxicity and confound chemical-biology studies. We report a novel series of small molecules based on a tricyclic pyrimidothiazolodiazepinone core that displays both high potency and selectivity for FAK. Structure-activity relationship (S AR) studies explored modifications to the thiazole, diazepinone, and aniline tail, which identified lead compound BJG-03-025. BJG-03-025 displays potent biochemical FAK inhibition (IC50 = 20 nM), excellent kinome selectivity, activity in 3D-culture breast and gastric cancer models, and favorable pharmacokinetic properties in mice. BJG-03-025 is a valuable chemical probe for evaluation of FAK-dependent biology.

Thiazolyl-pyrazoline derivatives: In vitro and in silico evaluation as potential acetylcholinesterase and carbonic anhydrase inhibitors

Alzheimer’s disease (AD) is a complex, predominant, and progressive form of dementia. The treatment of AD alters depending on the cognitive and behavioral symptoms. The utility of cholinergic replacement by acetylcholinesterase (AChE) inhibitors in AD treatment has been well-documented so far. Recent studies have also demonstrated that human carbonic anhydrases (hCAs) serve as important targets for AD treatment. In an attempt to identify potent AChE and hCA inhibitors, new thiazolyl-pyrazolines (3a-k) were designed based on the molecular hybridization of thiazole and pyrazoline scaffolds. A facile and versatile synthetic route consisting of three steps, namely Claisen-Schmidt reaction, the formation of the 2-pyrazoline ring system, and Hantzsch thiazole synthesis was used to prepare compounds 3a-k. The synthesized derivatives were experimentally validated for efficacy by in vitro and direct enzymatic assays. Furthermore, the compounds were subjected to in silico screening using Schrodinger Suite software to identify the binding affinities of potential compounds based on Glide XP scoring, MM-GBSA calculating, and validation. The results of in vitro and in silico studies revealed that compounds 3a, 3f, and 3d were the most promising derivatives in this series due to their significant effects on AChE, hCA I, and hCA II, respectively. Incorporation of Poly(Itaconic Acid) with Quaternized Thiazole Groups on Gelatin-Based Films for Antimicrobial-Active Food Packaging

Poly(itaconic acid) (PIA) was synthesized via conventional radical polymerization. Then, functionalization of PIA was carried out by an esterification reaction with the heterocyclic groups of 1,3-thiazole and posterior quaternization by N-alkylation reaction with iodomethane. The modifications were confirmed by Fourier transform infrared (FTIR) and proton nuclear magnetic resonance (H-1-NMR), as well as zeta-potential measurements. Their antimicrobial activity was tested against different Gram-negative and Gram-positive bacteria. After characterization, the resulting polymers were incorporated into gelatin with oxidized starch and glycerol as film adjuvants, and dopamine as crosslinking agent, to develop antimicrobial-active films. The addition of quaternized polymers not only improved the mechanical properties of gelatin formulations, but also decreased the solution absorption capacity during the swelling process. However, the incorporation of synthesized polymers increased the deformation at break values and the water vapor permeability of films. The antioxidant capacity of films was confirmed by radical scavenging ability and, additionally, those films exhibited antimicrobial activity. Therefore, these films can be considered as good candidates for active packaging, ensuring a constant concentration of the active compound on the surface of the food, increasing products’ shelf-life and reducing the environmental impact generated by plastics of petrochemical origin.

The novel sensitive and selective chemosensors for determination of multiple analytes

Novel chemosensors that are azo dyes bearing coumarin with indole and thiophene recognition moieties were synthesized. Sensor properties of the dyes were investigated within multiple basic environments that include various pH ranges, anions, and organic amines. Eleven adducts from 4-methylbenzo[d]thiazol-2-amineand Carboxylic Acids via Classical H-bonds and Noncovalent Associations

Cocrystallization of4-methylbenzo[d]thiazol-2-amine with an array of carboxylic acids got a total of 11 crystalline adducts. The 11 adducts have been examined by XRD, IR and EA, the melting points of all adducts were also gauged.Their structural and supramolecular aspects are fully analyzed. The result reveals that among the investigated crystalline solids1-4, 6-7 and 9-10 the aryl N in thiazole cores are protonated when the organic acids are deprotonated and the crystal packing is interpreted by the strong charge-assisted N-H center dot center dot center dot O H-bond from the NH+ and the carboxylates. 5, 8 and 11 are cocrystals sustained by the neutral N-H center dot center dot center dot O/O-H center dot center dot center dot N H-bonds. Except the N-H center dot center dot center dot O H-bonds, the O-H center dot center dot center dot OH-bonds were also present at 3, 6, 7 and 9-11. 1 has the additional N-H center dot center dot center dot S H-bonds.The O-H center dot center dot center dot S H-bond was created in 10. 1 and 5 contained the N-H center dot center dot center dot N H-bond. 4 showed the N-H center dot center dot center dot Cl H-bond.Further analysis of the crystal packing of the adducts told that a different set of additional O-H center dot center dot center dot C,C-C,O-C, O-O, O-S,Cl-O, Cl-Cl, Cl-S, Br-Br,CH3-N, CH-O/CH2-O/CH3-O, CH3-Cl/CH-Cl, C-pi, CH3-CH3 CH3 -CH/CH-CH, NH-pi, CH3 -pi/CH2 -pi/CH- pi, Cl-pi and pi-pi – contacts contribute to the stabilization and expansion of the totalstructures. For the interplay of the various weak nonbonding contacts these structures took on homo/hetero supramolecular synthons.The recognition of the base-acid in all the multicomponent crystalsis based on thetypical R-2(2) (8) synthon.
