The technique was designed primarily to monitor real-time changes in the concentration of known volatiles during eating and has limited identification power. However, for reliable quantification, it is necessary to assign the ions monitored in-nose with the volatile compounds present in the food. Besides assignments made on nominal mass, isotopic ratio analysis and accurate mass measurements have also been used to determine the contribution of certain compounds to a particular ion intensity. Discrimination between defective and non-defective Brazilian coffee beans by their volatile profile

The coffee roasted in Brazil is considered to be of low quality, due to the presence of defective coffee beans that depreciate the beverage quality. In view of the fact that coffee flavour is directly related to the volatile compounds produced during roasting, the objective of the present study was to perform a comparative evaluation of the volatile fraction of defective (black, immature, sour) and healthy coffee beans, in order to find possible chemical markers for detection of defective coffee beans in roasted coffee. Volatiles extraction and concentration was performed by solid phase micro-extraction (SPME) of the roasted coffee headspace, using a triple phase (divinylbenzene/carboxen/polydimethylsiloxane) fiber. Analysis of the volatile profiles was performed by GC-MS. The results obtained showed that the proposed methodology was adequate for extraction, concentration and analysis of the coffees volatile profile. Several substances were identified as possible markers for differentiating black, sour and immature beans from healthy coffee beans. Statistical analysis of the data by principal components (PCA) demonstrated that the volatile profile enables the differentiation of healthy and defective coffees. The data were separated into two major groups, one represented by immature and black beans and the other by healthy and sour coffee beans. Such results indicated that black and sour beans can be associated to fermentation of immature and of healthy beans, respectively.

Singlet Oxygen Photooxidation of Peptidic Oxazoles and Thiazoles

Oxazoles and thiazoles are commonly found moieties in nonribosomal peptides (NRPs) and ribosomally synthesized post-translationally modified peptides (RiPPs), which are important biomolecules present in the environment and in natural waters. From previous studies, they seem susceptible to oxidation by singlet oxygen (1O2); therefore, we designed and synthesized model oxazole- and thiazole-peptides and measured their1O2 bimolecular reaction rate constants, showing slow photooxidation under environmental conditions. We reasoned their stability through the electron-withdrawing effect of the carboxamide substituent. Reaction products were elucidated and support a reaction mechanism involving cycloaddition followed by a series of rearrangements. The first1O2 bimolecular reaction rate constant for a RiPP, the thiazole-containing peptide Aerucyclamide A, was measured and found in good agreement with the model peptide’s rate constant, highlighting the potential of using model peptides to study the transformations of other environmentally relevant NRPs and RiPPs.

Champagne regulations allow winegrowers to stock still wines to compensate for quality shifts in vintages, mainly due to climate variations. According to their technical requirements and house style, Champagne producers use these stored wines in their blends to enhance complexity. The presence of lees and aging at low pH (2.95-3.15), as in Champagne wines, lead to several modifications in wine composition. These conditions, combined with extended aging, result in the required environment for the Maillard chemical reaction, involving aromatic molecules, including sulfur, oxygen, and nitrogen heterocycles (such as thiazole, furan, and pyrazine derivatives), which may have a sensory impact on wine. Some aromatic heterocycles in 50 monovarietal wines aged from 1 to 27 years provided by Veuve Clicquot Ponsardin Champagne house were determined by the SPME-GC-MS method. The most interesting result highlighted a strong correlation between certain heterocycle concentrations and wine age. The second revealed a correlation between heterocyclic compound and free amino acid concentrations measured in the wines, suggesting that these compounds are potential aromatic precursors when wine is aged on lees and, thus, potential key compounds in the bouquet of aged Champagnes. The principal outcome of these assays was to reveal, for the first time, that aromatic heterocycle concentrations in Champagne base wines are correlated with wine age.
