Known and novel histaminergic H1- and H2-receptor agonists were investigated as potentially potent and selective brain-penetrating compounds. While no novel compound was identified which satisfied our requirements for a brain-penetrating agonist, betahistine 14 and 2-(thiazol-2-yl)ethylamine 16 can be regarded as H1-receptor agonists with moderate brain-penetrating ability, of potential value as pharmacological tools. A novel histamine analogue, N,N-bis-{2-[4(5)-imidazolyl]ethyl}amine 25 is reported which, although unlikely to be brain penetrant, was found to be equipotent with histamine at H1- and H1-receptors.

The results allow fitting of parameters in the Hamiltonian that describes internal rotation of the CH3 group about its local C3 axis. The V3 terms in the periodic potential energy functions that describe the internal rotation in N-, 2-, 4-, and 5-methylimidazole are determined to be 185.104(11), 122.7529(38), 317.20(14), and 386.001(19) cm-1, respectively, by the internal axis method. The experiments are supported by density functional theory calculations. Observed variations in barrier height are explained with reference to the symmetry of overlap between a pi-like orbital on the CH3 group and pi-orbitals on the aromatic ring.

A new heteroleptic Ir(I) compound exhibiting high volatility and defined thermal decomposition under CVD conditions is reported. The new iridium precursor [(COD)Ir(ThTFP)] (COD = cyclooctadiene, ThTFP = (Z)-3,3,3-trifluoro-1-(thiazol-2-yl)prop-1-en-2-olate) unifies both reactivity and sufficient stability through its heteroleptic constitution to provide a precise control over compositional purity in CVD deposits. The solution integrity of the monomeric Ir(I) complex was investigated by 1D and 2D NMR spectroscopy and EI mass spectrometry, whereas the molecular structure was confirmed by single-crystal diffraction. CVD experiments demonstrated the suitability of the iridium compound for an atom-efficient (high molecule-to-precursor yield) gas-phase deposition of nanocrystalline iridium films that could be converted into crystalline iridium dioxide upon heat treatment to demonstrate their electrocatalytic potential in the oxygen evolution reaction.
